Purpose:We evaluated the efficacy and safety of Celecoxib 400 mg capsules taken orally for 12 weeks in patients with osteoarthritis of the knee. Patients were assigned to one of three treatment groups (n = 60), divided into 3 treatment groups (n = 60), each containing 200 mg of Celecoxib 400 mg capsules. Group 1 (n = 20) received Celecoxib 400 mg capsules twice daily for 12 weeks; group 2 (n = 20) received Celecoxib 400 mg capsules once daily for 12 weeks. Group 3 (n = 30) received Celecoxib 400 mg capsules twice daily for 12 weeks. Patients received either 50 mcg or 200 mcg of Celecoxib 400 mg capsules once daily for 12 weeks.
Methods:We used a 2-week double-blind phase II study. Patients (N = 60) were enrolled between December 2014 and January 2015. Patients were randomized to one of three treatment groups: Celecoxib 400 mg capsules (group 1, n = 20) given by mouth for 12 weeks, Celecoxib 400 mg capsules (group 2, n = 20) given by oral intramuscular injection for 12 weeks, or placebo (placebo, n = 30). Patients were given either 50 mcg or 200 mcg of Celecoxib 400 mg capsules once daily for 12 weeks.
Results:The incidence of adverse events was significantly higher in the Celecoxib 400 mg capsules (n = 20) group than in the placebo (n = 20) group. The incidence of the adverse events in group 1 was significantly higher than in group 2 (n = 20). The frequency of discontinuation of the therapy was similar between groups. The incidence of gastrointestinal (GI) adverse events was significantly higher in the Celecoxib 400 mg capsules (n = 20) group than in the placebo (n = 20) group (3.5% versus 0.4%, respectively).
Conclusions:Celecoxib 400 mg capsules were more efficacious than Celecoxib 400 mg capsules for 12 weeks.
